Australia will play New Zealand on October 25 at WIN Stadium in Wollongong, followed by Tonga at Eden Park on November 2.
Klemmer, who played in 17 Tests during his six seasons with Canterbury, will be Newcastle's first Kangaroo since Dane Gagai in 2017.
He is the first Newcastle front-rower chosen for Australia since club legend Paul Harragon in 1998.
Meanwhile, dynamic Knights fullback Kalyn Ponga has been named in the Australian squad for the World Nins tournament at Bankwest Stadium on October 18-19.
Newcastle trio Hannah Southwell, Holli Wheeler and Isabelle Kelly were named in the Jillaroos side to take on the Silver Ferns in the curtain-raise to the Kangaroos v Kiwis Test.
