Doyle, who is now based at Newcastle, claimed the $150,000 Muswellbrook Cup (1500 metres) with eight-year-old Petrology.
On Saturday, Pride Of Adelaide will contest the group 2 Tulloch Stakes at Rosehill for Doyle, who grew up in Muswellbrook.
On Friday, Petrology, which came to Doyle from Matthew Dunn midway through last year, settled near last before jockey Ashley Morgan found a gap at the top of the straight.
Petrology, which last won in June 2017 at Moonee Valley, burst to the lead with 200m left and went on to win by more than three lengths.
It was Doyle's biggest win to date and came after Petrology started his preparation with two sixth-placed efforts in town. Friday's win was his seventh in 62 starts.
"I'd just like to thank Mark Pilkington for giving me a go with this horse," Doyle told Sky Thoroughbred Central.
"He had a couple of runs, coming to us from another trainer at the back end of last preparation, and I just wanted to tip him out and start fresh, and I said to Mark that I had this cup in mind.
"We gave him two runs with no blinkers, but he was set for today and what a great ride by Ash, too. He comes in and rides a lot of work for us and this old horse runs for him and he just rode him a peach today."
Pride Of Adelaide is a $5 chance on Saturday.
"He's fit and well and I'm happy with that horse, and we're ready to have a crack at them," Doyle said.
