The chopper, four ambulance crews and nine SES rescue operators responded to single car accident at Nelsons Plains where a car rolled and a woman was trapped.
The car left the road and became airborne before crashing and rolling several times across a nearby field. A NSW Ambulance spokesperson said the 29-year-old woman was trapped for "quite a while" after the crash on Seaham Road, which occurred about 11pm.
Port Stephens SES were called in to help with the rescue which they described as "a complex and extended extrication".
The roof of the car had to be peeled away and several sides of the vehicle were removed so that paramedics could treat the patient.
The woman was flown to John Hunter Hospital with leg and pelvis injuries in a serious condition.
Earlier in the day, the chopper and three ambulance crews responded to an incident at Raworth about 3.30pm where a 32-year-old man came off a motorcycle on Ellie Avenue.
The man suffered a head injury and grazes to his body. He was taken to John Hunter Hospital by ambulance in a serious condition.
