Chevy Royale has won her past four starts, and nine of 17 in her career, but faces a rise in class from box four in the second heat, where the top two progress to the final.
She won from box six last week in her Wentworth Park debut, running 29.8 seconds.
"She went all right there the other day," Manuel said.
"She missed the start a fraction and the two kept her wide through the first bend. She checked off its heels a little bit coming off the back straight then cut to the fence and away she went, so I think there's a little bit of improvement in her, but it's whether it's enough to beat that class of dog.
"There's a few that are better than handy. Vicki's Queen in the one is pretty experienced around there and is coming off the Easter Egg, so it's a massive jump for us to go from fifth grade to that."
Also Saturday, The Gardens has a 12-race program from 5.04pm.
