The left-handed pitcher is part of the extended bench for the Baltimore Orioles, who are playing against the Toronto Blue Jays at New York's Sahlen Field on Saturday (AEST).
Baltimore made the announcement after going down 9-0 to Toronto in game one of their series on Friday.
The clubs face off again in back-to-back encounters on Sunday and Monday.
Wells was promoted from triple-A side Norfolk along with right-handed pitcher Konner Wade.
"I want to have a healthy season and pitch in the big leagues," Wells told the Newcastle Herald a few months ago.
"I want to go over there and compete and hopefully find myself up in the active 25-man roster and make my debut.
"It's the thing I've been dreaming of since I started playing baseball so to tick that off as a milestone in my life would be good."
Belmont Baseball Club, where Wells started his career, released a statement on social media on Saturday.
"Well it's old boys 'Back 2 Belmont' weekend and what better way to celebrate than to have our very first MLB player," the club representative said.
"Congratulations to Alexander Wells on your call up to the Baltimore Orioles MLB team.
"Belmont Baseball Club wish you all the very best and are very proud of your continued achievements.
"Congratulations to Kylie, Darren and family as well. It's going to be a great weekend and we will watch with immense pride."
