Explosive Venom has won her past three starts, twice at The Gardens over 515m and once at the Maitland 565m, but she returns to the 600m for the $5000-to-the-winner GRNSW Middle Distance Series race on the 12-event card.
Her only attempt over the track's 600m was on July 10 when 4.25 lengths second to the Steven Burgis-trained Cawbourne Canelo.
They meet again on Saturday night but this time Explosive Venom has the better draw in three and Cawbourne Canelo the six.
Explosive Venom has also posted best-of-the-night times at each of her past three wins.
"She's flying," Flanagan said. "She's a different bitch now we've sorted her box manners. Straight after that [600m] run, we started doing the box work with her, and bang, away she went.
"We've just changed a few things up with her and she's running the times we thought she could.
"I'm thinking she'll just jump, go to the lead and that will be it tomorrow night. Once she gets out in front, she takes over and she has that strong finish. I'd say she'll be too strong for the others."
On Friday at The Gardens, Richmond Vale trainer Jason Mackay had a winning treble with Straight Shift, Zipping Eve and Zipping Maserati.
Mark Davidson-trained Mickey Doo scorched to a 12-length win in 29.33 seconds for the 515m in his third consecutive victory.
Harry Sarkis (All Most There and Impress Scarlett) had a double.
