A newly-diagnosed case from Sydney "was associated with the Costco fuel station and the external membership office", the heath district advised on Monday afternoon.
Anyone who attended Costco fuel station or the onsite membership office between Monday and Friday last week (inclusive) is urged to get tested today or tomorrow and isolate until their result is returned.
The instruction includes people who have already been tested since the exposure came to light on Friday.
"The outbreak at Costco has grown to a total of six known cases (some of who reside in Sydney), with transmission on site indicating a strong likelihood that there will be more cases," Hunter New England Health said in a statement.
"The risk associated with this venue has been reassessed following the new information we have received, which is why we are requesting causal contacts have an additional test."
Both Sunday and Monday's update included one case linked to the Costco exposure, which had more than 400 associated contacts.
