The outside back has been the club's top try-scorer in each of the past two seasons, after coming from North Queensland.
He is now signed to the club until the end of the 2023 season.
Tuala has missed just one game in the two seasons with Newcastle, scoring 24 tries.
Knights head of recruitment Clint Zammit believed the 23-year-old had untapped potential.
"Enari worked hard on the things he needed to change, he played in every game for the Knights in 2021, resulting in a career-best season," Zammit said in a statement.
"His extension adds real depth to our outside backs, covering both wing and centre.
"If he continues to work hard, especially in the pre-season, we believe his game can improve again 2022."
The news comes as the Knights search for a replacement for experienced halfback Mitchell Pearce, who was granted a release from the final year of his contract at Newcastle to take up a three-year-old deal with Catalans Dragons.
