Police were called to New Line Road about 5.50pm on Sunday after a Mazda sedan left the road, hit a power pole and began to roll.
The driver and front-seat passenger, a 29-year-old woman and three-year-old girl respectively, suffered facial lacerations.
They were taken to the John Hunter Hospital for treatment.
A six-year-old boy who had been travelling in the back seat died at the scene.
The driver has been subjected to mandatory blood and urine tests.
A crime scene was established at the scene and a report will be prepared for the information of the coroner.
Specialists from the Metropolitan Crash Investigation Unit are continuing inquiries into the circumstances of the crash.
