The group, which is featured regularly on Triple J radio, includes brother and sister Joel and Naomi Wenitong and Jacob Turier (DJ Jaytee).
The trio have been touring this month with fellow hip hop artist Urthboy (aka Tim Levinson) on the back of the release of their album Next of Kin.
No stranger to being recognised at the indigenous awards, The Last Kinection claimed their third consecutive Deadly award last year for best hip-hop group.
This year best hip-hop group went to Melbourne-based duo the Yung Warriors.
Jessica Mauboy was named female artist of the year while Gurrumul Yunupingu picked up male artist of the year.
The film Mabo, which aired on the ABC1 in June, was the big winner of the night claiming best flick, best actress (Deborah Mailman) and best actor (Jimi Bani).
NRL Bulldogs fullback Ben Barba, who won the 2012 Dally M Medal, scored the Deadly for NRL player of the year.
The national Aboriginal and Torres Strait Islander music, sport, entertainment and community awards were held at the Sydney Opera House.
Hosted by singer Casey Donovan and actor Luke Carroll, this was the 18th year of the awards.
The Deadlys will be broadcast on SBS1 on Sunday .
