The store is part of the $35 million Mayfield East Village shopping centre, which also includes a bottle shop, specialty stores and 250 underground car parking spaces.
The development has been in the works for more than a decade with the initial plans approved in 2010.
The designs were refined multiple times over the ensuring years and a squabble about nearby road upgrades delayed the project until it gained final approval in 2019.
Coles Group chairman James Graham cut the ribbon on the new store alongside former Newcastle Knights player and Man Shake founder Adam MacDougall on Wednesday.
Coles has employed 112 people to work at the supermarket, which multiple existing Mayfield traders recently said would be a boost for the suburb.
Store manager Amber Gourley said the store would play an important role in the lives of locals for years to come and was the only supermarket in Mayfield with underground parking.
"After years of planning, we're proud to be opening the doors to Mayfield's newest supermarket destination, offering customers more choice, great value and the latest in-store supermarket features," she said.
"We've got new store features that Newcastle shoppers probably haven't seen before, including a pick-and-mix bar just for dogs and more than 100 convenient ready-made meal options."
Mayfield also has Woolworths and Aldi supermarkets.
