In front of a record 10,515 people, the hosts scored 14 unanswered points in the second half after trailling by four at the main break.
Blues halfback and player of the match Maddie Studdon, who scored the third and final try for NSW with nine minutes remaining, said she felt "over the moon" with the victory.
Maroons five-eighth Ali Brigginshaw expressed her "disappointment" with the result.
Queensland crossed the line in the 14th minute but the try was disallowed.
Cameron Park's Hannah Southwell also produced a try-saving tackle under the goal posts for NSW.
The Maroons then opened the scoring in the 25th minute courtesy of second-rower Tazmin Gray, who dummied and stepped back inside the Blues defence for a 4-0 lead.
Shortly after half-time NSW had a try turned down.
Following a penalty awarded to Wallsend's Holli Wheeler near halfway, the Blues levelled proceedings at 4-all in the 39th minute through right-edge winger Jessica Sergis.
Six minutes later opposite winger Shakiah Tungai put NSW ahead 8-4 on the left-hand side of the field after five-eighth Kirra Dibb retained possession via a 40-30 kick.
Studdon sealed the triumph with a solo effort next to the sticks, converting her own try.
"It was really good seeing so many people come out and support us, the atmosphere was unreal," Blues centre, CRL Newcastle player and 2018 Golden Boot recipient Isabelle Kelly said.
"The first 30 minutes was such a grind, but in the second half we stuck it to them and got the win."
Earlier at the same venue on Friday, Newcastle's Caitlin Johnston was named player of the match in the inaugural women's under-18 clash with NSW prevailing 24-4 over Queensland.
