Family members, friends and lord mayor John Tate will unveil the tribute near the former Lynch's Prawns site.
The site marks the western end of Joy Cummings Promenade, which stretches to the pilot station in the east.
Cr Tate said the late Mrs Cummings had made a significant contribution to Newcastle.
"Joy was chosen as the first female lord mayor in Australia [in 1974]," Cr Tate said.
"This is a significant achievement and I [am] honoured to unveil a plaque commemorating Joy's contribution to our city."
The site recognises her role, during her time on the council, in the transformation of the foreshore into a recreation area.
She said she had always hoped the city would be opened to the harbour.
