Johnston, born in Belmont and aged 20, was unveiled by the club on Thursday ahead of Newcastle's maiden campaign in the new-look national women's competition later this year.
The prop previously played for the Knights in juniors, part of the club's foundation Tarsha Gale Cup team in 2018 followed by a season as captain in 2019.
She joins the Knights after two NRLW seasons with the Sydney Roosters.
"I felt having come through the pathways here at the Knights, it was time to come home," Johnston told the club's media.
"This gives our girls an opportunity to get where they want to in their rugby league careers and I'm really looking forward to it.
"I feel I'll bring my aggression and knowledge of the game to the team.
"I've been mixing it with the best-known players and I'm looking forward to leading from the front."
