The reigning Women's Premier League minor-major premiers arrive at the annual knockout tournament somewhat understrength, but just four wins away from claiming the top prize.
O'Brien, who had surgery on a ruptured Achilles tendon five weeks ago, says some of Olympic's regular first graders will be missing which gives other players an opportunity.
"We've only got about four or five first graders, but it's given the younger girls a chance to really step up," O'Brien said.
"We've got a lot of depth at our club. There's about 12 or so from reserves and maybe four from 17s getting a run. It's really exciting for them and not as much pressure as weekend fixtures. A few of the other girls have niggles so it's good for them to have some time off."
Jemma House and Georgia Amess are among those in action with Olympic drawn against Charlestown and Garden Suburb, who replaced Mayfield United this week. Kotara South have also pulled out, leaving 14 teams in total.
O'Brien has experienced State Cup success previously, lifting the trophy with Wallsend in 2019. Adamstown won in 2018 and Warners Bay picked up the inaugural crown. Last year's competition was cancelled because of coronavirus.
University's Chelsea Nesbitt, who joined the club this year after relocating from Albury, will meet Cooks Hill, Mid Coast and Broadmeadow. The Students are eligible for the Community Plate.
Play starts on Friday night. Preliminary rounds continue on Saturday. Semis and finals follow on Sunday.
